Abstract | This study aims to obtain wide genetic base and new juice cane clone as specific area. There were 4 steps at the first step so callus was induced from young leaf Suphanburi 50 Juice cane so its were cultured on modified MS (Murashige and Skoog, 1962) with 2,4-D (dichlorophenoxyacetic acid) 5 mg/l and 10 % coconut juice. Its were kept at dark and control temperature condition room. Mutation Inducing using callus cultured in modified MS media with Sodium azide (NaN3) 5 concentration as 0, 5, 10, 15 and 20 mg/l, the second step. Then, there were induced adventitious shoot from callus mutant in the third step. The last step, inducing root respectively. Mutation breeding using NaN3 mutagen resulted in calluses from young leaf after cultured 8 weeks and increased callus by more than 6 times subculture. Then, there were callus cultured on modified MS media with 5 mg/l NaN3. Its was survival and mass increased in during time. After that, the adventitious shoot developed after callus cultured 5 weeks and the most at 8 weeks. In addition so adventitious shoot were subculture for increasing shoot in the same media at 4 to 5 weeks. Finally, adventitious shoot were induced on modified MS at the after cultured 2 weeks. |
