งานวิจัยชิ้นนี้มีวัตถุประสงค์เพื่อคัดแยกแบคทีเรียที่ได้จากตัวอย่างดินที่ได้จากเขตอุทยานแห่งชาติน้ำหนาว ในบริเวณจังหวัดเพชรบูรณ์ และชัยภูมิ และคัดแยกแบคทีเรียที่สามารถสร้างสารออกออกฤทธิ์ทางชีวภาพ (bioactive compound) และหาสภาวะที่เหมาะสมต่อการสร้างสารออกฤทธิ์ทางชีวภาพ ผลที่ได้จากการแยกเชื้อแบคทีเรียจากดินในอุทยานน้ำหนาว สามารถแยกได้ 10 ไอโซเลต เมื่อนำมาเพาะเลี้ยงลงในอาหารเลี้ยงเชื้อแบบจำเพาะในแต่ละสภาวะเพื่อหาสภาวะที่เหมาะสมเพื่อเหนี่ยวนำให้มีการสร้างสารทุติยภูมินั้นพบว่ามี 2 ไอโซเลตได้แก่ S1 และ S2 ที่สามารถสร้างสารทุติยภูมิ
ได้ดีในอาหาร Glucose yeast peptone medium นำอาหารเหลวที่ได้จากการเพาะเลี้ยงมาสกัดแยกโดยวิธี liquid-liquid extraction ด้วย ไดคลอโรมีเทนได้สารสกัด S1DCM และ S2DCM และสกัดด้วยเอทธิลอะซีเตทได้สารสกัด S1OAC และS2OAC นำสารสกัดหยาบที่ได้ในแต่ละชั้นไปทดสอบความเป็นพิษกับเซลล์มะเร็งลำไส้ชนิด HT-29 และ HCT116 ที่ความเข้มข้นตั้งแต่ 0.1-1.0 มิลลิกรัมต่อมิลลิลิตรหาค่าร้อยละการตายของเซลล์ด้วยวิธี MTT assay และ cell apoptosis assay พบว่าสารสกัดหยาบที่นำมาทดสอบนั้นมีความสามารถในการยับยั้งการเจริญของเซลล์มะเร็ง HT-29 และ HCT116 ได้ดีโดยเฉพาะสารสกัด S2OAC ให้ค่าการยับยั้งเซลล์มะเร็งร้อยละ 50 ที่ความเข้มข้น 0.5 มิลลิกรัมต่อมิลลิลิตร และพบว่าทำให้เกิดการตายในระยะเริ่มต้นและระยะสุดท้ายของเซลล์มะเร็งชนิด HT-29 และ HCT116 เท่ากับร้อยละ 70 และ 80 ตามลำดับ โดยสรุปสารทุติยภูมิที่ได้จากแบคทีเรียไอโซเลต S1 และ S2 ในตัวอย่างดินสามารถนำไปพัฒนาเพื่อใช้เป็นสารต้านมะเร็งลำไส้ได้ต่อไป

 Abstract
The objective of this research is to isolate bacteria from soil samples obtained from Nam Nao National Park. In Phetchabun and Chaiyaphum provinces, and screening bacteria that are capable of produce bioactive compounds and finding suitable conditions for the production of bioactive substances. The results of bacterial isolation from the soil in the Nam Nao National Park can be separated into 10 isolates, but when cultured in a specific culture medium in each condition to find suitable conditions to induce the product bioactive compound, it was found that 2 isolates S1 and S2 could produce secondary metabolite in the Glucose yeast peptone medium. The liquid-liquid extracted from culture was extracted by a partition with dichloromethane (S1DMC, S2DMC), ethyl acetate (S1OAC, S2OAC) and the aqueous layer and evaporate until dry before leading to the in vitro assay. Colorectal cells including HT-29 and HCT116 were incubated with S1OAC, S2OAC, S1DCM, and S2DCM extracts at the dose of 0.1-1.0 mg/ml for 24 h and cell viability was evaluated by MTT assay. The extracts inhibited cell proliferation in HT29 and HCT116 cells in a dose dependent manner with S2OAC had highest inhibitory effect at concentration at 50% (IC50) approximately 0.5 mg/ml in both cells compared with the control cells treated with 0.1% DMSO. Following treatment of extract S2OAC at 1 mg/ml for 24 h on HT-29 and HCT116, cancer cells turned into the early and late stages of apoptosis approximately at 70% and 80% respectively. It suggests apoptosis effect following extract S2OAC treatment in colorectal cancer cells. The bacteria isolates of S1 and S2 which are isolate from soil sample from Nam Nao National Park can be further used as anticancer agents against colon cancer cell lines
